Prions

چکیده
plants are cloaked in hundreds of tiny projections, known as trichomes. These structures vary in function, shape, size and spacing from species to species, and it’s not known whether similar cellular differentiation programs are responsible for making trichomes in different plants. Trichomes are clearly visible in the scanning electron micrograph of a wild-type Nicotiana tabacum seedling above (bottom left). Petal epidermis is textured by different projections, called floral papillae, which are shown above at middle left. When the transcription factor MIXTA (which regulates papillae in Antirrhinum majus) is over-expressed in Nicotiana, petal papillae elongate to produce trichome-like structures and seedling leaves produce extra trichomes, as shown in the right-hand images. Although these specialized epidermal cells have nearly opposite functions — floral papillae attract pollinators by enhancing pigment perception, whereas leaf trichomes probably discourage hungry insects — these results suggest that, in Nicotiana, similar pathways regulate their development. Resistance of Bovine Spongiform Encephalopathy (BSE) Prions to Inactivation
Distinct prion strains often exhibit different incubation periods and patterns of neuropathological lesions. Strain characteristics are generally retained upon intraspecies transmission, but may change on transmission to another species. We investigated the inactivation of two related prions strains: BSE prions from cattle and mouse-passaged BSE prions, termed 301V. Inactivation was manipulated...
متن کاملAmyloid Prions in Fungi.
Prions are infectious protein polymers that have been found to cause fatal diseases in mammals. Prions have also been identified in fungi (yeast and filamentous fungi), where they behave as cytoplasmic non-Mendelian genetic elements. Fungal prions correspond in most cases to fibrillary β-sheet-rich protein aggregates termed amyloids. Fungal prion models and, in particular, yeast prions were ins...
متن کاملTransmission barriers for bovine, ovine, and human prions in transgenic mice.
Transgenic (Tg) mice expressing full-length bovine prion protein (BoPrP) serially propagate bovine spongiform encephalopathy (BSE) prions without posing a transmission barrier. These mice also posed no transmission barrier for Suffolk sheep scrapie prions, suggesting that cattle may be highly susceptible to some sheep scrapie strains. Tg(BoPrP) mice were also found to be susceptible to prions f...
متن کاملTransgenetic studies implicate interactions between homologous PrP isoforms in scrapie prion replication.
Transgenic (Tg) mice expressing both Syrian hamster (Ha) and mouse (Mo) prion protein (PrP) genes were used to probe the mechanism of scrapie prion replication. Four Tg lines expressing HaPrP exhibited distinct incubation times ranging from 48 to 277 days, which correlated inversely with HaPrP mRNA and HaPrPC. Bioassays of Tg brain extracts showed that the prion inoculum dictates which prions a...
متن کاملPrions Ex Vivo: What Cell Culture Models Tell Us about Infectious Proteins
Prions are unconventional infectious agents that are composed of misfolded aggregated prion protein. Prions replicate their conformation by template-assisted conversion of the endogenous prion protein PrP. Templated conversion of soluble proteins into protein aggregates is also a hallmark of other neurodegenerative diseases. Alzheimer's disease or Parkinson's disease are not considered infectio...
متن کاملPrions, protein homeostasis, and phenotypic diversity.
Prions are fascinating but often misunderstood protein aggregation phenomena. The traditional association of the mammalian prion protein with disease has overshadowed a potentially more interesting attribute of prions: their ability to create protein-based molecular memories. In fungi, prions alter the relationship between genotype and phenotype in a heritable way that diversifies clonal popula...
